Hey Mike,

It completely depends on what BIOS you have, and if the BIOS settings for your HDD's are set to "Auto". In many cases, yes, you would need to enter BIOS.

No motherboard settings would need to be changed. Are you planning on a clean install with your new HDD?

If you find that it does not detect your HDD and you need to change BIOS, this is quite easy. Press [DEL] or [F1] at the initial post screen to enter setup, and let me know the name and version of BIOS that is displayed across the top, and I can talk you through it.

Would it be possible to use my old 2.5" hard drive as extra storage on my desktop PC if so is there a cable to attatch it to a USB port?

Of course. If you wanted it to attach it to USB, you would need a hard drive caddy, like this one: NexStar 3 USB 2.0 HDD Enclosure

However, your PC will normally allow up to 4 internal IDE devices (CD-ROM's, HDD's). So if your not using all 4, then you can keep it internal. Look for a spare 4 pin power plug in your PC, and a free port on the ribbon cable. If you can't see any spare, purchasing these and keeping your HDD internal would be cheaper than buying a caddy.
