I have a Dell Lattitude laptop and I just hooked up an external enclosure with another hard drive, via usb. The system recognizes it and the usb controller, but I can't access it through "my computer" or windows explorer. Am I doing something wrong or am I missing something?

If your hard drive is new one then your need to create the partition first so, to create the partition of your hard drive , right click on the my computer and select the manage option and click on the disk management and it will show all the hard drive whichever plugged with your machine then select the your new hard drive and format it, then create the partition and it will show in the my computer. some times virus infection some hard doesn't show in the my computer or open.
