I tested this on Windows Media Centre, which went as far as to say that it was not detecting an audio device. This is odd, because when I view connected hardware it does detect my speakers, and they do produce sound.
Well, this does change things - in the sense that we're not looking at a problem localized to Flash. Do you have any yellow exclamation marks in Device Manager?

Yes, it's interesting that it would work in certain cases using Windows Media Player, but not others.

The only yellow exclamation mark that appears in the Device Manager is a network adapter called the Microsoft 6to4 Adapter.

That being the case, though, any ideas on how to fix this?

It's just that it's been over a week - and I do have a video project I'm working on that's expected to be done by next month.

Just a bit of an update. I don't think I've mentioned this, but there is still no audio when headphones are plugged in to the computer. Thought this might be a helpful tidbit, because it shows that the problem isn't actually the speaker hardware.

If you're having trouble with Adobe Flash Player, it must be completely removed before re-installation. Read the removal instructions carefully and follow them to the letter:

How to uninstall the Adobe Flash Player plug-in and ActiveX control

Please note that you should save the downloaded file to your desktop as you will have to close every running program before removal. All I can tell you is this proceedure has worked for me in the past.

Thank you for the advice. However, in the past couple of pages, we've established that it's not a problem localized in Flash Player as it's been discovered in a few other programs as well.
