Hi JHE, and welcome to PC Help Forum.

If either the keyboard or mouse are PS2 (small round connections) you will have to reboot the computer for them to reinitialize properly. If they are USB (small rectangular connection) you should have no problems. USB is plug and play and are meant to be able to be inserted and removed while the computer is on. I wouldn't recommend doing it still unless necessary as sometimes the silliest little things can wreak havok on your operating system. Not to say that it will but why take the chance. If you have to remove the keyboard and/or mouse it shouldn't cause any physical damage to the peripherals but just because something shouldn't happend also doesn't necessarily mean it won't happen.

I wouldn't unplug a PS2 connection when the pc is on anyway.

Only USB devices and my headset , nothing else when the pc is on.
